Intended Use:
Ideal for staining bacterial or fungal smears, enabling differentiation between gram-positive and gram-negative microorganisms through microscopic examination.
How to Use:
Smear Preparation: Prepare a bacterial smear (avoid excessive density), air-dry, and fix by passing through a flame 1-2 times. Ensure the slide remains cool to the touch during fixation.
Primary Staining: Apply Reagent A to cover the smear, stain for 60 seconds, rinse with water, and blot dry.
Mordanting: Apply Reagent B to cover the smear, stain for 60 seconds, rinse with water, and blot dry.
Decolorization: Apply Reagent C, decolorize for 15-30 seconds, rinse with water, and blot dry.
Counterstaining: Apply Reagent D to cover the smear, stain for 60 seconds, rinse with water, and blot dry.
Microscopy: Observe under an oil immersion lens.
Result Interpretation:
Gram-positive bacteria appear blue to blue-purple.
Gram-negative bacteria appear magenta to red.
Performance Indicator:
Staining Effect: Pseudomonas aeruginosa stains red (rod-shaped), and Bacillus subtilis stains blue (rod-shaped) under microscopy.
