
Geobacter metallireducens Medium (ATCC Medium 1768) | Basal Medium for Geobacter Cultivation
Geobacter metallireducens Medium (ATCC Medium 1768) is a basal medium designed for the cultivation of Geobacter metallireducens. This medium requires supplementation with ferric citrate solution and sodium acetate solution for optimal growth under anaerobic conditions.

Formula (per liter):
Wolfe’s Vitamin Solution – 10 ml
Wolfe’s Mineral Solution – 10 ml
NaHCO₃ – 2.5 g
NH₄Cl – 0.25 g
NaH₂PO₄·H₂O – 0.6 g
KCl – 0.1 g
Preparation Instructions:
Dissolve 3.51 g of medium in 950 ml of distilled or deionized water, boil gently until fully dissolved.
Dispense into suitable containers.
For every 95 ml of medium, aseptically add 1 vial of Ferric Citrate solution and 1 vial of Sodium Acetate solution.
Cool to 25℃.
Adjust pH to 5.0 with sterile 10N NaOH and maintain stability.
Sparge with an N₂/CO₂ (80:20) gas mixture to create anaerobic conditions.
Adjust final pH to 6.8–7.0.
Sterilize by autoclaving at 121℃ for 15 minutes.
