Gauze’s Synthetic Broth Medium is a classical liquid medium formulated for the cultivation of Actinomycetes. It provides soluble starch as the main carbon source and essential mineral salts to support microbial growth. This medium is suitable for laboratory research and microbiological studies focusing on Actinomycetes.

Medium Composition (per liter):

  • Soluble Starch: 20.0g

  • Potassium Nitrate (KNO3): 1.0g

  • Dipotassium Phosphate (K2HPO4): 0.5g

  • Magnesium Sulfate (MgSO4): 0.5g

  • Ferrous Sulfate (FeSO4): 0.01g

  • Sodium Chloride (NaCl): 0.5g

pH adjusted to 7.3 ± 0.1 at 25°C.

Instructions for Use:

  1. Dissolve 22.5g of medium in 1L distilled or deionized water.

  2. Heat to boiling for at least 1 minute until fully dissolved.

  3. Adjust final volume to 1000ml with water, dispense, and sterilize at 121°C for 15 minutes.

  4. After cooling, optional: for every 300ml medium, add 1ml of 3% potassium dichromate solution to increase selectivity. Mix gently before use.

Storage:
Store at room temperature. Shelf life: 3 years.
