- Pancreatic Digest of Casein ……………………. 2.5 g
- Peptic Digest of Animal Tissue ……………….. 2.5 g
- Bile Salts ……………………………………………….. 1.0 g
- Calcium Carbonate ………………………………… 10.0 g
- Sodium Thiosulfate …………………………………. 30.0 g
⚗️ Preparation Instructions
- Suspend 46.0 g of dehydrated medium in 1,000 ml of distilled or deionized water
- Heat to boiling with stirring
- Dispense into suitable containers
- Sterilize by autoclaving at:
- 121 °C for 15 minutes, or
- 115 °C for 30 minutes
- Cool and store the sterilized base in a cool, dark place
Supplementation Before Use
- Before use, add one vial of iodine solution and one vial of 0.1% brilliant green solution to each 100 ml of prepared medium
- Mix thoroughly while gently shaking
- Dispense immediately while mixing
🧫 Storage & Stability
- Store dehydrated medium at room temperature, protected from light and moisture
- Supplemented medium should be used according to laboratory protocols
⚠️ Limitations & Precautions
- Supplements must be added only after sterilization and cooling
- Do not use medium showing caking, discoloration, or moisture absorption
- Not intended for clinical diagnostic use
