EF-18 Agar Base is a selective medium designed for membrane filtration testing of Salmonella species. It is formulated with sulfapyridine, yeast extract, and L-lysine to enrich the growth of Salmonella while inhibiting the growth of competing microorganisms. The medium contains bile salts to further enhance its selective properties. Each 100 ml of prepared medium requires the addition of Novobiocin solution to ensure selectivity and prevent non-target bacterial growth.

Composition (per liter)

  • Proteose Peptone: 5.0 g

  • Yeast Extract: 3.0 g

  • L-Lysine Monohydrochloride: 10.0 g

  • Dextrose: 2.5 g

  • Sucrose: 15.0 g

  • Magnesium Sulfate: 1.5 g

  • Bile Salts: 1.5 g

  • Sulfapyridine: 0.3 g

  • Bromthymol Blue, Sodium Salt: 0.03 g

  • Agar: 15.0 g

  • Final pH: 7.1 ± 0.2 at 25℃

Preparation Instructions

  1. Dissolve 5.38 g of medium in 100 ml of distilled or deionized water by boiling for 1 minute until fully dissolved.

  2. Cool the solution to approximately 50°C.

  3. Add 1.5 mg of Novobiocin solution per 100 ml of medium and mix well.

  4. Pour into sterile Petri dishes to solidify. Store in a cool, dry place for future use.
