Dubos Oleic Agar Base is a selective medium designed for the isolation and cultivation of Mycobacterium tuberculosis. When used with Dubos Oleic Agar Base Additive and penicillin, this medium supports optimal growth of tuberculosis-causing bacteria. The formula contains casein digest, asparagine, essential salts, and agar to maintain nutrient balance and stable pH.

Composition (per liter)

  • Pancreatic Digest of Casein – 0.5g

  • Asparagine – 1.0g

  • Monopotassium Phosphate – 1.0g

  • Disodium Phosphate (anhydrous) – 2.5g

  • Ferric Ammonium Citrate – 0.05g

  • Magnesium Sulfate – 0.01g

  • Calcium Chloride – 0.5mg

  • Zinc Sulfate – 0.1mg

  • Copper Sulfate – 0.1mg

  • Agar – 15.0g

  • pH 6.6 ± 0.2 (25℃)

Preparation Instructions

  1. Dissolve 2.0g of Dubos Oleic Agar Base in 90ml distilled or deionized water and heat until completely dissolved.

  2. Sterilize at 121℃ for 15 minutes.

  3. After sterilization, shake gently to prevent agar sedimentation and cool to approximately 55℃.

  4. Aseptically add 10ml Dubos Oleic Agar Base Additive (2 vials) and 2,500–5,000 U penicillin (0.5–1 vial penicillin solution).

  5. Mix gently and pour plates for cultivation.

Notes

  • The powder may settle or clump, but this does not affect medium performance.

  • Ensure aseptic technique when adding additives and penicillin.
