Product Overview:
Dnase Test Agar is a specialized medium used for the detection of deoxyribonuclease (DNase) activity in microorganisms. It is widely applied in microbiological laboratories for the identification and differentiation of DNase-positive bacteria.
Formula (per liter):
Deoxyribonucleic Acid – 2.0 g
Pancreatic Digest of Casein – 15.0 g
Papaic Digest of Soybean Meal – 5.0 g
Sodium Chloride – 5.0 g
Agar – 15.0 g
pH 7.3 ± 0.2 (25℃)
Preparation Instructions:
Weigh 42.0 g of the medium and suspend in 1 L of distilled or deionized water.
Heat to boiling with constant stirring until completely dissolved.
Dispense into suitable containers.
Sterilize at 121℃ for 15 minutes.
Mix well immediately after sterilization to prevent agar from settling.
Test Method:
Inoculate and incubate at 35℃ for 18–24 hours.
After incubation, flood the agar surface with 1 mol/L hydrochloric acid solution.
A clear zone around colonies indicates DNase-positive reaction.
Alternatively, add 0.1% toluidine blue solution: unhydrolyzed DNA turns blue, while hydrolyzed areas appear rose-pink.
