Test Protocol:
1. Suspend 42.0g in 1L distilled water
2. Boil with stirring until completely dissolved
3. Autoclave at 121°C for 15 minutes
4. Inoculate and incubate at 35°C for 18-24 hours
5. Flood with 1M HCl or 0.1% toluidine blue solution
Result Interpretation:
– With HCl: Clear zones around DNase-positive colonies
– With toluidine blue: Blue (negative) vs pink (positive)
