Formula Composition (per liter):
– Pancreatic Digest of Casein: 5.0g
– Glucose: 10.0g
– Soybean Lecithin: 7.0g
– Sodium Thiosulfate: 6.0g
– Sodium Hydrogensulfite: 2.5g
– Yeast Extract: 2.5g
– Sodium Thioglycollate: 1.0g
– Bromcresol Purple: 0.02g
– Final pH: 7.6 ± 0.2 (25°C)
Preparation Protocol:
1. Suspend 34.0g in 1L distilled water
2. Heat to 80°C with vigorous stirring
3. Add 1g Tween 80 per 200ml broth
4. Autoclave at 121°C for 15 minutes
Note: Clumping during suspension is normal – continue heating to dissolve
Quality Validation:
– Neutralization capacity tested against benzalkonium chloride
– Growth recovery ≥70% with Pseudomonas aeruginosa
– Low background bioburden
