Composition (per liter)

  • Potato Powder: 5.0g
  • Dextrose (Glucose): 20.0g
  • Monopotassium phosphate (KH₂PO₄): 3.0g
  • Magnesium Sulfate (MgSO₄): 1.5g
  • Thiamine•HCl (Vitamin B1): 0.1g
  • Agar: 20.0g

Final pH: 6.0 ± 0.2 at 25°C

Instructions for Preparation

  1. Suspend 49.6g of the powder in 1 liter of distilled or deionized water.
  2. Heat and boil to dissolve the medium completely.
  3. Dispense into final containers.
  4. Sterilize by autoclaving at 121°C for 15 minutes or 115°C for 30 minutes.
  5. After sterilization, gently swirl the vessel to prevent agar sedimentation before the medium solidifies.
  6. Cool and prepare slants, plates, or broths as required.
