Columbia Blood Agar Base is a dehydrated culture medium base used for the preparation of blood agar plates by supplementing with sterile defibrinated sheep blood.
It is widely applied in microbiological laboratories for the cultivation of fastidious organisms, including beta-hemolytic streptococci and Campylobacter species.

The medium provides a nutritionally rich foundation with excellent buffering capacity and consistent gel strength, supporting reliable colony development and hemolysis observation after blood supplementation.

🧾 Medium Composition (per liter)

  • Peptic Digest of Animal Tissue – 23.0 g

  • Sodium Chloride – 5.0 g

  • Starch – 1.0 g

  • Agar – 15.0 g

Final pH: 7.2 ± 0.2 (25 °C)


⚙️ Directions for Use

  1. Suspend 44.0 g of the medium in 1 L of distilled or deionized water.

  2. Heat with stirring and boil for at least 1 minute until completely dissolved.

  3. Dispense as required and sterilize by autoclaving at 121 °C for 15 minutes.

  4. Cool the medium to 45–50 °C.

  5. Under aseptic conditions, add 5 mL of sterile defibrinated sheep blood (pre-warmed to 35 °C) per 100 mL of medium.

  6. Mix gently and pour into sterile Petri dishes.
