Overview
Columbia Blood Agar Base is a high-quality culture medium widely used for the cultivation of bacteria, particularly fastidious microorganisms. When supplemented with sterile defibrinated sheep blood, it provides a nutrient-rich environment suitable for isolation and growth of demanding bacterial species. It is commonly applied in clinical, research, and diagnostic microbiology laboratories.

Pancreatic Digest of Casein: 10.0 g
Yeast Extract: 5.0 g
Peptic Digest of Meat: 5.0 g
Beef Heart Digest: 3.0 g
Sodium Chloride: 5.0 g
Corn Starch: 1.0 g
Agar: 15.0 g
pH: 7.3 ± 0.2 (at 25°C)
Preparation Instructions
Weigh 44.0 g of Columbia Blood Agar Base powder and add to 1 L of distilled or deionized water.
Heat with stirring and boil for at least 1 minute until fully dissolved.
Dispense into suitable containers.
Sterilize by autoclaving at 121°C for 15 minutes or at 115°C for 30 minutes.
Cool to 45–50°C.
Under aseptic conditions, add 5–7% sterile defibrinated sheep blood (pre-warmed to 35°C).
Mix gently and pour into plates for use.
