Application

CAS Assay Medium is designed for the screening and detection of bacterial siderophores. It is widely used in microbiology and environmental research to evaluate iron-chelating activity.

Composition (per liter)

• Chrome Azurol S (CAS) – 60.5 mg

• Hexadecyltrimethylammonium bromide (HDTMA) – 72.9 mg

• FeCl3·6H2O – 2.645 mg

• Agar – 9.0 g

• 0.1 mol/L Phosphate Buffer – 50 mL

Phosphate Buffer Content (per 50 mL):

• NaH2PO4·2H2O – 295.25 mg

• Na2HPO4·12H2O – 1213.5 mg

• NH4Cl – 125 mg

• KH2PO4 – 37.5 mg

• NaCl – 62.5 mg

pH: 6.8 ± 0.2 (25℃)

Instructions for Use

1. Weigh 10.87 g of the medium and dissolve in 1 L of distilled or deionized water.

2. Heat to boiling for at least 1 minute until dissolved.

3. Dispense into containers and autoclave at 115℃ for 20 minutes.

4. Shake well after sterilization.

5. Cool to 50–55℃ before use.

⚠️ Note: The medium may contain insoluble substances after preparation, which is normal.
