Charcoal Agar (Regan-Lowe Charcoal Agar Medium) is designed for the selective cultivation of aerobic bacteria, particularly Bordetella pertussis (whooping cough). This medium is charcoal-enriched and free from nicotinic acid to promote the growth of specific bacteria in clinical and research applications. It’s ideal for isolating Bordetella pertussis and other aerobic pathogens.

Instructions for Use:

  1. Dissolve 62.5g of Charcoal Agar in 1L of distilled or deionized water.

  2. Heat and dissolve completely.

  3. Sterilize by autoclaving at 121°C for 15 minutes or 115°C for 30 minutes.

  4. After sterilization, gently shake the medium to prevent agar from settling.

  5. If needed, cool to 50°C, then add 1 vial of Cefalexin solution (100mg) and 10ml sterile defibrinated sheep blood per 100ml medium.

  6. Gently mix and pour onto plates.

Storage: Store at room temperature, away from direct sunlight. Shelf life: 3 years.

  • Beef Heart Infusion: 12.0g

  • Peptone: 10.0g

  • Sodium Chloride (NaCl): 5.0g

  • Soluble Starch: 10.0g

  • Yeast Extract: 3.5g

  • Charcoal Bacteriological: 4.0g

  • Agar: 18.0g

pH: 7.3 ± 0.2 at 25°C
