Chapman Stone Agar Medium
Chapman Stone Agar Medium is a microbiological culture medium formulated for the isolation and cultivation of Staphylococcus aureus.
The medium contains high salt concentration and mannitol, which create selective conditions suitable for the growth of Staphylococcus species while inhibiting many other microorganisms.

Ammonium Sulfate ((NH₄)₂SO₄) ………. 75.0 g
D-Mannitol ……………………………… 10.0 g
Sodium Chloride ………………………… 55.0 g
Dipotassium Phosphate (K₂HPO₄) ……. 5.0 g
Gelatin ………………………………….. 30.0 g

Preparation Instructions
- Suspend 202.5 g of the medium in 1 liter of distilled or deionized water.
- Heat and boil for approximately 1 minute until the medium is completely dissolved.
- Adjust the volume to 1000 ml if necessary.
- Dispense into suitable containers.
- Sterilize at 121°C for 15 minutes or 116°C for 30 minutes.
- After sterilization, mix well to prevent agar from settling at the bottom before solidification.
- Cool to approximately 55°C before use.
Storage
Store tightly sealed in a cool and dry place at room temperature.
