
Bushnell-Haas Broth | Hydrocarbon Utilization Test Medium for Bacteria
Bushnell-Haas Broth is a selective medium formulated without cholesterol, primarily used for the cultivation of Nocardia species and for studying the microbial utilization of hydrocarbons. This medium provides essential inorganic salts and requires the addition of a hydrocarbon source to evaluate microbial biodegradation capacity.

Formula (per liter):
Potassium Dihydrogen Phosphate – 1.0 g
Dipotassium Hydrogen Phosphate – 1.0 g
Ammonium Salt – 1.0 g
Magnesium Sulphate·7H₂O – 0.2 g
Ferric Chloride – 0.05 g
Calcium Chloride·2H₂O – 0.02 g
Final pH: 7.2 ± 0.2 (25℃)
Preparation Instructions:
Suspend 3.27 g of the medium in 1 liter of distilled or deionized water.
Heat to boiling until completely dissolved.
Dispense into suitable containers.
Sterilize by autoclaving at 121℃ for 15 minutes.
Mix well after sterilization.
For hydrocarbon utilization studies, aseptically supplement with 0.1–1.0% sterile hydrocarbon source after sterilization.
