Medium Composition
Composition per liter:
- Casein Peptone — 10.0 g
- Lactose — 10.0 g
- Sucrose — 10.0 g
- Sodium Chloride — 5.0 g
- Yeast Extract — 1.5 g
- Phenol Red — 0.08 g
- Crystal Violet — 0.002 g
- Agar — 14.0 g
Preparation Instructions
Mix gently and pour plates.
Suspend 50.58 g of the medium in 1 liter of distilled or deionized water.
Heat to boiling and maintain boiling for at least 1 minute until fully dissolved.
Dispense into suitable containers.
Sterilize by autoclaving at 121°C for 15 minutes.
Immediately mix well after sterilization.
Cool to 50–55°C.
Under aseptic conditions, add 1 vial of BCSA supplement per 200 ml of medium.
