Buffered Charcoal Yeast Extract Agar Base without L-cysteine (BCYE-Cys Agar Base) is a dehydrated culture medium designed for the isolation and identification of Legionella species.
It provides a selective medium without L-cysteine, making it suitable for differential testing and confirmatory diagnosis of Legionella in microbiological laboratories.

  • Activated Charcoal ……………………………. 2.0 g

Preparation and Usage

  1. Suspend 38.0 g of the medium in 1 L distilled or deionized water.

  2. Heat with frequent agitation until completely dissolved.

  3. Dispense in 100 ml volumes and autoclave at 121 °C for 15 minutes.

  4. Shake gently after sterilization and allow to cool to 50 °C.

  5. Aseptically add 1 vial of soluble ferric pyrophosphate per 100 ml of medium and mix well.

  6. Pour into sterile Petri dishes under aseptic conditions.

  7. Adjust pH with sterile KOH solution if necessary.
