Product Application:
Buffered Charcoal Yeast Extract Agar Base (BCYE) is used for the selective isolation and cultivation of Legionella species. It provides essential nutrients and buffering capacity to support the growth of Legionella while suppressing unwanted microorganisms. With supplementation, it can also be prepared as BCYE differential agar for enhanced colony identification.

Medium Formula (per liter):

  • Activated Charcoal – 2.0 g

  • Yeast Extract – 10.0 g

  • ACES Buffer – 10.0 g

  • α-Ketoglutarate – 1.0 g

  • Agar – 15.0 g

  • pH 6.85 ± 0.2 (25℃)

Instructions for Use:

  1. Weigh 38.0 g of the base medium and dissolve in 1 L distilled or deionized water.

  2. Boil until fully dissolved, dispense 100 ml per bottle.

  3. Sterilize at 121℃ for 15 minutes. Shake well after sterilization.

  4. Cool to approx. 50℃ and, under aseptic conditions, add the following supplements per 100 ml:

    • L-Cysteine Hydrochloride solution

    • Ferric pyrophosphate solution

  5. For BCYE differential agar, also add BCYE differential supplement.

  6. Adjust pH with KOH solution if necessary.

  7. Pour into plates and allow to solidify.
