Brilliant Green Phenol Red Agar (BGPA) is a selective and differential culture medium formulated for the isolation of Salmonella species from clinical, food, and environmental specimens. The medium contains brilliant green dye, which selectively inhibits Gram-positive bacteria and many Gram-negative organisms, while phenol red serves as a pH indicator to detect carbohydrate fermentation.

Medium Composition (per liter)

  • Peptone – 10.0 g

  • Beef Extract – 5.0 g

  • Yeast Extract – 3.0 g

  • Disodium Hydrogen Phosphate – 1.0 g

  • Monosodium Phosphate – 0.6 g

  • Lactose – 10.0 g

  • Sucrose – 10.0 g

  • Phenol Red – 0.09 g

  • Brilliant Green – 0.0125 g

  • Agar – 15.0 g

  • Final pH: 7.0 ± 0.2 (25°C)

Preparation Instructions

  1. Suspend 54.7 g of the medium powder in 1 liter of distilled or deionized water.

  2. Heat with frequent agitation until completely dissolved.

  3. Dispense into suitable containers.

  4. Sterilize by autoclaving at 121°C for 15 minutes (or at 115°C for 30 minutes).

  5. Mix thoroughly after sterilization to avoid agar sedimentation.

  6. Allow to solidify before use.
