Overview
Blood Agar Base is a high-quality culture medium used for the preparation of blood agar plates. When supplemented with sterile defibrinated sheep blood, it supports the growth of a wide variety of microorganisms, including fastidious bacteria. This medium is widely applied in clinical, research, and diagnostic microbiology laboratories for bacterial isolation, cultivation, and hemolysis observation.
Composition (per liter)
Peptone: 10.0 g
Beef Extract: 3.0 g
Sodium Chloride: 5.0 g
Agar: 15.0 g
pH: 7.3 ± 0.1 (at 25°C)
Preparation Instructions
Weigh 33.0 g of Blood Agar Base powder and add to 1 L of distilled or deionized water (adjust quantities proportionally if preparing a different volume).
Heat to boiling until fully dissolved.
Dispense into appropriate containers.
Sterilize by autoclaving at 121°C for 15 minutes or at 115°C for 30 minutes.
Shake gently after sterilization to prevent agar settling.
Cool to 50–55°C.
Under aseptic conditions, slowly add 5–7% sterile defibrinated sheep blood (pre-warmed to 35°C) along the inner wall of the container.
Mix gently and pour into plates; allow to solidify before use.
