BL Agar Base – Selective Medium for Bifidobacterium with Sterile Defibrinated Horse Blood

BL Agar Base is a specialized medium designed for the isolation and cultivation of Bifidobacterium species. This medium requires the addition of sterile defibrinated horse blood to support the growth of bifidobacteria. The formula includes L-Cysteine Hydrochloride and Tween 80 to enhance anaerobic growth and maintain medium stability.

Composition (per liter):

• Meat Extract – 2.4 g

• Proteose Peptone – 10.0 g

• Peptone – 5.0 g

• Soya Peptone – 3.0 g

• Yeast Extract – 5.0 g

• Liver Extract – 3.2 g

• Dextrose – 10.0 g

• Soluble Starch – 0.5 g

• KH2PO4 – 1.0 g; K2HPO4 – 1.0 g

• MgSO4 – 0.2 g; FeSO4 – 0.01 g

• NaCl – 0.01 g; MnSO4 – 0.007 g

• Bubble Absorbing Agent (Silicon) – 0.2 g

• Polysorbate 80 – 1.0 g

• L-Cysteine Hydrochloride – 0.5 g

• Agar – 15.0 g

• pH 7.2 ± 0.2 (25℃)

Preparation Instructions:

1. Dissolve 58 g of powder in 1 L of distilled or deionized water.

2. Heat to boiling until fully dissolved.

3. Sterilize at 115℃ for 20 minutes.

4. Shake well immediately after sterilization and cool to 50℃.

5. Gently add 5% sterile defibrinated horse blood (pre-warmed to ~35℃) and mix carefully.

6. Pour plates under aseptic conditions.
