BCG Skim Milk Medium – Selective Medium for Lactic Acid Bacteria

BCG (Bromcresol Green) Skim Milk Medium is designed for the selective cultivation of lactic acid bacteria (LAB). The medium contains skim milk and bromocresol green to indicate acid production during LAB growth.

Composition (per liter):

• Skim Milk – 100.0 g

• Yeast Extract – 10.0 g

• Bromocresol Green – 0.016 g

• Agar – 15.0 g

• pH 6.8 ± 0.2 (25℃)

Preparation Instructions:

1. Dissolve 12.5 g of powder in 100 mL of distilled or deionized water.

2. Sterilize at 121℃ for 15 minutes.

3. Shake well after sterilization to prevent agar sedimentation.

4. Cool to approximately 55℃, pour into plates, and allow to solidify.

Notes:

• The medium is ideal for monitoring acid production of LAB.

• Bromcresol green serves as a pH indicator, changing color in response to acid formation.
