Baar’s Medium is designed for the cultivation of sulfate-reducing bacteria, such as Desulfovibrio and Desulfotomaculum, typically used for studying sulfur-reducing processes. This medium contains essential components like Magnesium Sulfate, Yeast Extract, and Ferrous Ammonium Sulfate to promote the growth of these specialized microorganisms.

Formulation (Per Liter of Medium):

  • Magnesium Sulfate (MgSO₄·7H₂O): 2.0g

  • Potassium Hydrogen Phosphate (K₂HPO₄): 1.0g

  • Calcium Sulfate (CaSO₄): 1.0g

  • Ammonium Chloride (NH₄Cl): 0.5g

  • Yeast Extract: 1.0g

  • Ferrous Ammonium Sulfate (Fe(NH₄)₂(SO₄)₂): 0.5g

Usage Instructions:

  1. Dissolve 5.0g of Baar’s Medium in 1L of distilled or deionized water at a mild temperature.

  2. Add 1 vial (35% Sodium Lactate Solution) to every 100ml of the medium.

  3. Adjust the pH to 7.5 ± 0.2 using Sodium Hydroxide or Dilute Hydrochloric Acid.

  4. Sterilize the medium by autoclaving at 121°C for 15 minutes.

  5. After cooling to room temperature, the medium is ready for use.
