Modified Baar’s Medium with 2.5% NaCl is specifically designed for the cultivation of sulfate-reducing bacteria, such as Desulfovibrio and Desulfotomaculum. This formulation includes a higher concentration of NaCl (2.5%) to enhance bacterial growth and activity in saline environments, making it ideal for sulfate reduction studies.

Formulation (Per Liter of Medium):

  • Sodium Chloride (NaCl): 25.0g

  • Sodium Citrate (C₆H₅Na₃O₇): 5.0g

  • Magnesium Sulfate (MgSO₄): 2.0g

  • Calcium Sulfate (CaSO₄): 1.0g

  • Ammonium Chloride (NH₄Cl): 1.0g

  • Yeast Extract: 1.0g

  • Ferrous Ammonium Sulfate (Fe(NH₄)₂(SO₄)₂): 1.0g

Usage Instructions:

  1. Dissolve 36.0g of Modified Baar’s Medium in 1L of distilled or deionized water, heat and boil until completely dissolved.

  2. Sterilize by autoclaving at 121°C for 15 minutes.

  3. After sterilization, cool the medium to 55°C.

  4. Add 1 vial of 35% Sodium Lactate Solution (contains 0.35g Sodium Lactate and 0.05g Disodium Phosphate) to every 100ml of the medium.

  5. Adjust the pH to 7.5 at 25°C using sterile Sodium Carbonate Solution or Diluted Hydrochloric Acid.

  6. Mix thoroughly and the medium is ready for use.
