Azotobacter Broth (Mannitol) is a liquid medium formulated for the cultivation of nitrogen-fixing bacteria from soil samples. Mannitol serves as the primary carbon source to support the selective growth of Azotobacter species. This broth is ideal for enrichment and liquid culture studies of nitrogen-fixing microorganisms in soil microbiology research.

Formula (per liter):
Dipotassium Phosphate – 1.0 g
Magnesium Sulphate – 0.2 g
Sodium Chloride – 0.2 g
Mannitol – 20.0 g
Ferrous Sulphate – 0.005 g
Final pH: 8.3 ± 0.2 (25℃)
Preparation Instructions:
Suspend 21.4 g of the medium in 1 liter of distilled or deionized water.
Warm gently to dissolve completely.
Dispense into appropriate containers.
Add 1 vial of Soil Extract Solution per 100 ml of medium.
Sterilize by autoclaving at 121℃ for 15 minutes.
Cool to room temperature before use.
