Azospirillum Medium with Agar – 250g for Azospirillum Counting & Cultivation

Azospirillum Medium with Agar is a specialized solid growth medium designed for the cultivation and counting of Azospirillum, a nitrogen-fixing bacterium used in agricultural applications. This medium supports the growth of Azospirillum species, providing all the necessary nutrients for optimal bacterial activity. The inclusion of agar ensures the solidification of the medium for easier handling during microbial counting.

Ingredients Per Liter:

• Yeast Extract: 0.05g

• K2HPO4 (Dipotasium Phosphate): 0.25g

• FeSO4·7H2O (Iron Sulfate): 0.01g

• Na2MoO4·2H2O (Sodium Molybdate): 0.001g

• MnSO4·H2O (Manganese Sulfate): 0.002g

• MgSO4·7H2O (Magnesium Sulfate): 0.2g

• NaCl (Sodium Chloride): 0.1g

• CaCl2·2H2O (Calcium Chloride): 0.02g

• (NH4)2SO4 (Ammonium Sulfate): 1.0g

• Biotin (Vitamin B7): 0.0001g

• Bromothymol Blue: 0.025g

• D-Glucose (Glucose): 5.0g

• Agar: 15.0g

• pH: 7.1 ± 0.2 (at 25°C)

Instructions for Use:

1. Dissolve 21.66g of Azospirillum Medium in 1L of distilled or deionized water.

2. Heat and boil the mixture until fully dissolved.

3. Autoclave the medium at 121°C for 15 minutes to sterilize.

4. After sterilization, gently shake the medium to ensure uniformity.

5. Under sterile conditions, add 1 vial of 20% Malic Acid Solution (5ml per 200ml medium), mix well.

6. Pour the medium into Petri dishes and allow to solidify.

7. Ready for use in counting and cultivating Azospirillum bacteria.

Storage:

Store in a cool, dry place, away from direct sunlight. Ensure the container is tightly sealed to maintain the integrity of the medium.
