Azospirillum Broth Medium – 250g for Azospirillum Cultivation

Azospirillum Broth Medium is a liquid culture medium designed for the cultivation of Azospirillum, a nitrogen-fixing bacterium vital for agricultural processes. This medium contains essential nutrients and is formulated for optimal growth and maintenance of Azospirillum species in laboratory conditions.

Ingredients Per Liter:

• Yeast Extract: 0.05g

• K2HPO4 (Dipotasium Phosphate): 0.25g

• FeSO4·7H2O (Iron Sulfate): 0.01g

• Na2MoO4·2H2O (Sodium Molybdate): 0.001g

• MnSO4·H2O (Manganese Sulfate): 0.002g

• MgSO4·7H2O (Magnesium Sulfate): 0.2g

• NaCl (Sodium Chloride): 0.1g

• CaCl2·2H2O (Calcium Chloride): 0.02g

• (NH4)2SO4 (Ammonium Sulfate): 1.0g

• Biotin (Vitamin B7): 0.0001g

• Bromothymol Blue: 0.025g

• D-Glucose (Glucose): 5.0g

• pH: 7.1 ± 0.2 (at 25°C)

Instructions for Use:

1. Dissolve 6.66g of Azospirillum Broth Medium in 1L of distilled or deionized water.

2. Heat and dissolve completely.

3. Autoclave at 121°C for 15 minutes.

4. After sterilization, gently shake the solution to ensure uniformity.

5. Under sterile conditions, add 1 vial of 20% Sodium Malate Solution (5ml per 200ml medium).

6. Mix thoroughly and the medium is ready for use.

Storage:

Store the medium in a cool, dry place. Ensure the container is tightly sealed to prevent contamination.
