APA Medium Base is a specialized microbiological medium formulated for the cultivation of halophilic bacteria. With a high salt concentration and a rich blend of peptone, yeast extract, glucose, and minerals, this medium supports robust growth of salt-loving microorganisms. Instructions for Use:

  1. Dissolve 64.0g of APA Medium Base in 900ml distilled or deionized water.

  2. Boil for 1 minute until completely dissolved.

  3. Adjust final volume to 1L with distilled or deionized water.

  4. Sterilize by autoclaving at 121°C for 15 minutes.

  5. Cool to approximately 55°C, add 100ml of sterile 10% sodium carbonate solution, mix well.

  6. Adjust pH to 9.8 ± 0.2 (25°C). Medium is now ready for use.

Storage: Store at room temperature, away from direct sunlight. Shelf life: 3 years.

  • Sodium Chloride: 20.0g

  • Dextrose: 1.0g

  • Monopotassium Phosphate (KH2PO4): 2.0g

  • Magnesium Sulfate (MgSO4): 1.0g

  • Protease-peptone: 35.0g

  • Yeast Extract: 5.0g

  • pH: 9.8 ± 0.2 at 25°C
