Principle of the Method

This assay is based on the diffusion of antibiotics within an agar medium.
The diameter of the inhibition zone produced is related to the concentration or biological activity of the antibiotic.
Potency is determined by comparing inhibition zones produced by a reference standard and a test sample against a yeast indicator organism.

✅ Preparation Instructions

  1. Suspend 62.5 g of the product in 1,000 mL of deionized water.
  2. Heat with stirring until completely dissolved.
  3. Sterilize at 121°C for 15 minutes.
  4. Cool to 45–50°C and pour into sterile Petri dishes.
  5. Allow to solidify before use.

  • Inoculate the test microorganism into sterile agar cooled to 40–45°C.
  • Overlay evenly onto the surface of the base agar.
  • Allow 10–20 minutes at a temperature below optimal growth for antibiotic pre-diffusion.
  • Incubate at 25–30°C for 18–24 hours before reading results.
