Ampicillin MacConkey Agar Base is a selective medium designed for isolation of Aeromonas hydrophila from clinical, food, and environmental samples, combining bile salt inhibition with antibiotic selection.

Key Applications:

Supplement Requirement:

– Add 1 ampicillin vial per 100ml medium

Selective Components:

– Bile salts: Inhibit Gram-positive bacteria

– Ampicillin: Suppresses competing flora

– Lactose: Differentiates fermenters

Protocol:

1️⃣ Suspend 57.0g in 1L water

2️⃣ Boil 1 minute → Autoclave at 115°C for 15 minutes

3️⃣ Cool to 50-55°C

4️⃣ Add 1 ampicillin vial per 100ml

5️⃣ Mix gently → Pour plates

Typical Results:

A. hydrophila: Colorless, translucent colonies

– Lactose fermenters: Pink colonies
