Actinomycete Broth is a nutrient-rich medium formulated for the cultivation of Actinomycetes species. It contains essential protein digests, yeast extract, minerals, and carbohydrates to promote robust microbial growth. This medium can be prepared as broth, semi-solid, or solid agar depending on the application, making it suitable for microbiological research and laboratory studies.

Medium Composition (per liter):

  • Pancreatic Digest of Casein: 17.0g

  • Heart Muscle Infusion (solids): 2.0g

  • Yeast Extract: 10.0g

  • Sodium Chloride (NaCl): 5.0g

  • Dextrose (Glucose): 5.0g

  • Monopotassium Phosphate (KH2PO4): 13.0g

  • Monopotassium Phosphate (KH2PO4): 2.0g

  • Ammonium Sulfate ((NH4)2SO4): 1.0g

  • L-Cysteine HCl: 1.0g

  • Soluble Starch: 1.0g

  • Magnesium Sulfate (MgSO4): 0.2g

  • Calcium Chloride (CaCl2): 0.01g

pH adjusted to 7.3 ± 0.2 at 25°C.

Instructions for Use:
Dissolve 57.0g of medium in 1L of distilled or deionized water.

  • For semi-solid agar: add 7g/L agar.

  • For solid agar: add 20g/L agar.
    Heat until completely dissolved, dispense as required, and sterilize by autoclaving at 121°C for 10 minutes. Shake gently after sterilization to ensure uniform distribution.

Storage:
Store at room temperature. Shelf life: 3 years.
