7.5% Sodium Chloride Broth is a selective enrichment medium widely used for the isolation and homogenization of Staphylococcus aureus from food, clinical, and environmental samples. The high salt concentration (7.5% NaCl) inhibits most competing microorganisms while allowing the selective growth of staphylococci.

Composition (per liter):

  • Peptone – 10.0 g

  • Beef Extract – 5.0 g

  • Sodium Chloride – 75.0 g

  • Final pH: 7.4 ± 0.1 (at 25°C)


Preparation & Instructions:

  1. Suspend 90.0 g of the medium in 1 liter of distilled or deionized water.

  2. Warm gently to dissolve completely.

  3. Dispense into suitable containers.

  4. Sterilize by autoclaving at 121°C for 15 minutes.

  5. Allow to cool before use.
