🧾 Medium Composition (per liter)

  • Dehydrated Calf Brain Infusion – 4.0 g

  • Dehydrated Beef Heart Infusion – 4.0 g

  • Pancreatic Digest of Casein – 16.0 g

  • Peptone – 5.0 g

  • Dextrose – 2.0 g

  • Sodium Chloride – 5.0 g

  • Disodium Phosphate (Na₂HPO₄) – 2.5 g

  • Agar – 15.0 g

Final pH: 7.4 ± 0.2 (25 °C)


⚙️ Directions for Use

  1. Suspend 53.5 g of the medium in 1 liter of distilled or deionized water.

  2. Heat with stirring and boil until completely dissolved.

  3. Dispense into suitable containers.

  4. Sterilize by autoclaving at 121 °C for 15 minutes.

  5. Mix gently after sterilization and pour into sterile Petri dishes.

  6. Allow to solidify before use.
