10% Sodium Chloride Tryptic Soy Broth (10% NaCl TSB) is a selective enrichment medium designed for the growth and detection of Staphylococcus aureus, especially in Most Probable Number (MPN) tests. The high salt concentration (10% NaCl) effectively suppresses the growth of competing bacteria, while the enriched nutrients in tryptic soy broth support the selective proliferation of staphylococci.

Composition (per liter):

  • Pancreatic Digest of Casein (Tryptone) – 17.0 g

  • Enzymatic Digest of Soybean Meal – 3.0 g

  • Sodium Chloride – 100.0 g

  • Sodium Pyruvate – 10.0 g

  • Dipotassium Phosphate – 2.5 g

  • Dextrose – 2.5 g

  • Final pH: 7.3 ± 0.2 (at 25°C)


Preparation & Instructions:

  1. Suspend 135.0 g of the medium in 1 liter of distilled or deionized water.

  2. Heat until completely dissolved.

  3. Dispense into appropriate containers.

  4. Sterilize by autoclaving at 121°C for 15 minutes.

  5. Cool and store until use.
