
Raffinose-Histidine Medium – Agar-Based Medium for Actinomycete Isolation, 100g
Raffinose-Histidine Medium is a selective agar-based medium designed for the isolation and cultivation of Actinomycetes. The combination of raffinose as a carbon source and histidine as a nitrogen source provides an enriched environment that supports actinomycete differentiation while inhibiting unwanted microbial growth.
Complete Formula (per liter):
Histidine: 0.5 g
Raffinose: 2.5 g
Potassium Phosphate Tribasic (K₂HPO₄·3H₂O): 1.0 g
Magnesium Sulfate Heptahydrate (MgSO₄·7H₂O): 0.5 g
Ferrous Sulfate Heptahydrate (FeSO₄·7H₂O): 0.01 g
Calcium Chloride (CaCl₂): 0.02 g
Agar: 20.0 g
Final pH: 7.2 ± 0.2 at 25°C
Instructions for Use:
Dissolve 24.5 g of medium in 1 liter of distilled or deionized water.
Heat to boiling for at least 1 minute until the agar is completely dissolved.
Dispense into suitable containers.
Sterilize at 121°C for 15 minutes.
Cool and store until use.
