
Gauze’s Synthetic Agar Medium, Modified 250g for Actinobacteria Cultivation
Gauze’s Synthetic Agar Medium, Modified, is designed specifically for the cultivation of Actinobacteria. It is a modification of the original Gauze’s Synthetic Agar Medium, widely used for research and biotechnological applications involving Actinobacteria species, including actinomycetes.
Composition per liter:
Soluble Starch: 20.0 g
Potassium Nitrate (KNO3): 1.0 g
Dipotassium Phosphate (K2HPO4): 0.5 g
Magnesium Sulfate (MgSO4): 0.5 g
Sodium Chloride (NaCl): 0.5 g
Agar: 15.0 g
Final pH: 7.3 ± 0.1 (25℃)
Preparation:
Weigh 37.5 g of the medium and dissolve it in 1 L of distilled water or deionized water. Boil for 1 minute to dissolve completely, then add water to make up to 1000 ml. Dispense into containers and sterilize at 121℃ for 15 minutes. Shake the medium after sterilization to prevent agar from settling at the bottom. If needed, cool to 55℃, add 1 ml of 3% Potassium Chromate solution per 300 ml of medium to enhance antimicrobial properties, mix thoroughly, and pour into sterile plates.
