Application

Modified COMBO Medium Base is formulated for the cultivation of cyanobacteria, cryptophytes, green algae, and diatoms. It provides balanced nutrients for algal growth in laboratory research and environmental studies.

Composition (per liter)

• NaNO3 – 0.85 g

• CaCl2·2H2O – 0.3676 g

• MgSO4·7H2O – 0.3697 g

• NaHCO3 – 0.126 g

• Na2SiO3·9H2O – 0.2842 g

• K2HPO4 – 0.00871 g

• H3BO3 – 0.24 g

• KCl – 0.0745 g

• pH 7.8 ± 0.2 (25℃)

Instructions for Use

1. Dissolve 2.32 g of the medium in 1 L of distilled water.

2. Heat to boiling for 1 minute.

3. Cool to 25–30℃, then add one ampoule each of the following supplements:

   • Modified COMBO Medium Additive A: Algal trace element solution

   • Modified COMBO Medium Additive B: Vitamin B12 solution

   • Modified COMBO Medium Additive C: Biotin solution

   • Modified COMBO Medium Additive D: Thiamine solution

   • Modified COMBO Medium Additive E: CR1 soil extract solution

4. Adjust pH to 7.8 ± 0.2 using HCl or NaOH.

5. Filter to remove insoluble substances, then sterilize by filtration.

6. Medium is ready for use.
