Nematode Growth Medium (NGM), also known as Nematode Growth Agar Medium, is a widely used solid medium for the cultivation of Caenorhabditis elegans (C. elegans) and other nematodes. It provides the essential nutrients and minerals required for nematode growth and development in laboratory research.

For optimal use, each 100 mL of prepared NGM should be supplemented with a set of NGM Medium Additives (cholesterol, magnesium sulfate, and potassium phosphate solution). Optional addition of streptomycin sulfate can enhance selectivity and reduce microbial contamination.

Formula (per liter):

  • Peptone – 2.5 g

  • Sodium Chloride – 3.0 g

  • Calcium Chloride – 0.111 g

  • Magnesium Sulfate – 0.12 g

  • Cholesterol – 0.005 g

  • Potassium Phosphate Monobasic – 3.4 g

  • Agar – 17.0 g

Note: Magnesium sulfate, potassium phosphate, and cholesterol are supplied separately as NGM Medium Additives.


Preparation Instructions:

  1. Suspend 22.6 g of powder in 1 L distilled or deionized water.

  2. Heat and boil for at least 1 minute until completely dissolved.

  3. Dispense into suitable containers.

  4. Sterilize by autoclaving at 121℃ for 15 minutes.

  5. Cool to 50–55℃.

  6. Add 1 set of NGM Medium Additives per 100 mL medium.

  7. Mix gently and dispense into sterile containers.

  8. (Optional) Add 0.03 g streptomycin sulfate per 100 mL medium to increase selectivity.

  9. The appearance of slight precipitates after additive supplementation is normal and does not affect performance.
