Bordet-Gengou Fluid Medium Base | Bordet-Gengou Broth Medium Base

Bordet-Gengou Fluid Medium Base is a pH 6.7, peptone-free liquid medium designed for the enrichment of Bordetella pertussis (whooping cough bacteria). The medium requires supplementation with glycerol, sterile defibrinated sheep blood, and optional antibiotics for selective cultivation. It provides an ideal environment for the growth of fastidious Bordetella species in microbiology research and diagnostic applications.

Formula (per liter):

• Potato Infusion (from 125 g potatoes) – 4.5 g

• Sodium Chloride – 5.5 g

• Final pH: 6.7 ± 0.2 (25℃)

Preparation Instructions:

1. Dissolve 10.0 g of the medium in 1 liter of distilled or deionized water.

2. Warm gently to dissolve completely.

3. Dispense into suitable containers.

4. Add 1 g glycerol per 100 ml of medium.

5. Sterilize by autoclaving at 121℃ for 15 minutes.

6. Cool to below 50℃.

7. Under aseptic conditions, add 15% sterile defibrinated sheep blood and mix well.

8. For selective enrichment, optionally add:

   • 50 U Penicillin per 100 ml or

   • 4 mg Cephalexin per 100 ml
