Medium Composition (per liter)

• Peptone: 15.0 g

• Sodium Chloride: 5.0 g

• Soluble Starch: 1.0 g

• Dipotassium Phosphate: 4.0 g

• Monopotassium Phosphate: 1.0 g

• Agar: 15.0 g

• Final pH: 7.4 ± 0.2 (25℃)

Preparation Instructions

1. Suspend 41.0 g of the medium in 1 L distilled or deionized water.

2. Heat to boiling for at least 1 minute until fully dissolved.

3. Dispense into suitable containers and sterilize by autoclaving at 121℃ for 15 minutes.

4. Cool to ~55℃, then aseptically add 7 ml sterile defibrinated sheep blood per 100 ml medium.

5. Mix thoroughly and use as required.
