Dubos Broth Base – 250g for Mycobacterium tuberculosis Cultivation
Dubos Broth Base is a selective medium designed for the cultivation of Mycobacterium tuberculosis. It is used with Dubos Broth Base Additive to support the growth of tuberculosis-causing bacteria. The medium contains casein digest, asparagine, Polysorbate 80, and essential salts to provide nutrients while maintaining a stable pH for optimal bacterial growth.

Pancreatic Digest of Casein – 0.5g
Asparagine – 2.0g
Polysorbate 80 – 0.2g
Monopotassium Phosphate – 1.0g
Disodium Phosphate (anhydrous) – 2.5g
Ferric Ammonium Citrate – 0.05g
Magnesium Sulfate – 0.01g
Calcium Chloride – 0.5mg
Zinc Sulfate – 0.1mg
Copper Sulfate – 0.1mg
pH 6.6 ± 0.2 (25℃)
Preparation Instructions
Dissolve 6.25g of Dubos Broth Base in 90ml distilled or deionized water at moderate temperature.
Sterilize at 121℃ for 15 minutes.
Cool to approximately 55℃.
Aseptically add 10ml of Dubos Broth Base Additive (2 vials) and mix gently.
The medium is ready for Mycobacterium tuberculosis cultivation.
Notes
The powder may clump due to Polysorbate 80, but this does not affect performance.
