Glucose Tryptone Agar is a selective culture medium designed for the enumeration of thermophilic bacterial spores. It provides a balanced composition of tryptone and glucose with an indicator dye for reliable microbial analysis.

Composition (per liter):

  • Tryptone – 10.0 g

  • Glucose – 5.0 g

  • Bromcresol Purple – 0.04 g

  • Agar – 15.0 g

  • Final pH: 6.7 ± 0.1 (25℃)

Preparation Instructions:

  1. Suspend 30.0 g of the medium in 1 L of distilled or deionized water.

  2. Heat to boiling until fully dissolved.

  3. Dispense into suitable containers.

  4. Sterilize at 121℃ for 20 minutes.

  5. Shake well after sterilization to avoid agar sedimentation.

  6. Cool and store until use.
