Plate Count Agar with BCP is a specialized agar medium formulated for enumeration of total lactic acid bacteria (LAB). The addition of Brom Cresol Purple (BCP) allows clear visualization of microbial growth, facilitating accurate colony counting in fermented products.

Composition (per liter):

  • Peptone – 5.0 g

  • Yeast Extract – 2.5 g

  • Dextrose – 1.0 g

  • Polysorbate 80 (Tween 80) – 1.0 g

  • L-Cysteine – 0.1 g

  • Brom Cresol Purple (BCP) – 0.06 g

  • Agar – 15.0 g

  • pH: 7.0 ± 0.2 (25℃)

Preparation Instructions:

  1. Dissolve 24.7 g of powder in 1 L of distilled or deionized water.

  2. Heat to boiling until fully dissolved.

  3. Dispense and sterilize by autoclaving at 121℃ for 15 minutes.

  4. After sterilization, mix thoroughly to prevent agar settling.

  5. Pour into plates and allow to solidify. Medium is ready for total LAB counting.
