Trypticase Soy Polymyxin Broth Base (TSPB) is a microbiological medium used for the Most Probable Number (MPN) test of Bacillus cereus. This broth base provides a reliable formulation for bacterial detection and enumeration in laboratory and research applications.

Medium Composition (per liter):

  • Pancreatic Digest of Casein 17.0g

  • Enzymatic Digest of Soybean Meal 3.0g

  • Sodium Chloride 5.0g

  • Dextrose 2.5g

  • Dipotassium Hydrogen Phosphate 2.5g

Preparation Instructions:

  1. Dissolve 30.0g of the medium in 1L of distilled or deionized water with gentle warming.

  2. Dispense 15ml into each test tube.

  3. Sterilize by autoclaving at 121°C for 15 minutes.

  4. After cooling below 50°C, aseptically add 0.3ml of S1302 per tube before use.

  5. Alternatively, after sterilization, add one vial of TSPB supplement per 100ml medium, mix well, and dispense into sterile tubes.
