Specialized Medium for Hydrogen Sulfide Production Testing
This base medium requires addition of lead acetate solution to detect black precipitate formation in bacterial H2S tests. Essential for differentiating Salmonella, Citrobacter, and other sulfide-producing pathogens.

Peptone 10.0g | Beef Extract 3.0g | NaCl 5.0g
Na₂S₂O₃ 2.5g | Agar 15.0g
▶ pH: 7.3±0.1 (25°C)
▶ Preparation:

  1. 35.5g/L → boil → 121°C/15min sterilization

  2. Cool 50-55°C → add 10ml 10% sterile lead acetate
