Cholera Double Sugar Iron Agar is a differential medium designed for comprehensive biochemical testing of Vibrio cholerae, combining dual sugar fermentation and hydrogen sulfide production tests in a single tube.

Protocol:

1️⃣ Prepare medium per instructions

2️⃣ Dispense into tubes

3️⃣ Autoclave and solidify as deep butt + slant

4️⃣ Stab inoculate butt and streak slant

5️⃣ Incubate 18-24 hours

Interpretation Guide:

– V. cholerae: Acid butt (yellow) / Alkaline slant (red) / No H₂S

– H₂S positive: Black precipitate
