Alkaline Agar is a selective medium for isolating Vibrio cholerae from clinical and environmental samples using pH-based inhibition.

Primary Application:

✔️ Cholera outbreak investigations

✔️ Water and food safety testing

✔️ Clinical specimen processing

Formula (Per Liter):

– Peptone: 10.0g

– Beef Extract: 3.0g

– Sodium Chloride: 5.0g

– Agar: 15.0g

pH 8.3-8.5 (25°C)

Protocol:

1️⃣ Suspend 33.0g in 1L water

2️⃣ Boil to dissolve

3️⃣ Autoclave at 121°C for 15 minutes

4️⃣ Swirl before pouring plates
