Plate Count Agar, also known as Standard Methods Agar or Tryptone Glucose Yeast Agar (TGYA), is a high-quality medium designed specifically for colony total count determination. This 250g package provides a reliable solution for laboratory use, ensuring consistent results in microbial enumeration.
Formulated to support optimal microbial growth, each liter of the medium contains key components that nourish microorganisms: Pancreatic Digest of Casein (5.0g) for nitrogen and amino acid supply, Yeast Extract (2.5g) for essential vitamins and growth factors, Glucose (1.0g) as an energy source, and Agar (15.0g) to form a solid gel matrix. The medium is adjusted to a pH of 7.0±0.2 at 25℃, creating an ideal environment for colony development.
Preparing the medium is straightforward:
Weigh 23.5g of the powder and dissolve in 1L of distilled or deionized water.
Heat and boil until fully dissolved.
Dispense into containers, then sterilize at 121℃ for 15 minutes or 115℃ for 30 minutes.
Shake well after sterilization to prevent agar sedimentation, and cool before use.
