The addition of neutral activated charcoal helps neutralize toxic metabolic byproducts, protecting delicate microorganisms during transit. Its optimized pH of 7.3 ± 0.1 at 25 °C supports specimen stability for reliable downstream testing.

Charcoal (neutral activated carbon): 10.0 g
Sodium chloride: 3.0 g
Sodium hydrogen phosphate: 1.15 g
Potassium dihydrogen phosphate: 0.2 g
Potassium chloride: 0.2 g
Sodium thioglycollate: 1.0 g
Calcium chloride: 0.1 g
Magnesium chloride: 0.1 g
Agar: 7.5 g
Preparation Instructions
Weigh 23.3 g of this medium and add to 1 L of distilled or deionized water.
Heat to boiling until fully dissolved, stir well.
Dispense into suitable containers.
Autoclave at 121 °C for 15 minutes.
After sterilization, cool to 50–55 °C, gently shake to ensure even distribution of activated carbon, then rapidly cool in cold water to solidify.
