Amies Transport Medium with carbon is formulated for the safe collection, transportation, and preservation of pathogenic bacterial specimens. The addition of neutral activated charcoal helps neutralize toxic metabolic byproducts, protecting delicate microorganisms during transit. Its optimized pH of 7.3 ± 0.1 at 25 °C supports specimen stability for reliable downstream testing.

Composition (per Liter)

• Charcoal (neutral activated carbon): 10.0 g

• Sodium chloride: 3.0 g

• Sodium hydrogen phosphate: 1.15 g

• Potassium dihydrogen phosphate: 0.2 g

• Potassium chloride: 0.2 g

• Sodium thioglycollate: 1.0 g

• Calcium chloride: 0.1 g

• Magnesium chloride: 0.1 g

• Agar: 7.5 g

Preparation Instructions

1. Weigh 23.3 g of this medium and add to 1 L of distilled or deionized water.

2. Heat to boiling until fully dissolved, stir well.

3. Dispense into suitable containers.

4. Autoclave at 121 °C for 15 minutes.

5. After sterilization, cool to 50–55 °C, gently shake to ensure even distribution of activated carbon, then rapidly cool in cold water to solidify.
